An anterograde transsynaptic AAV1 carrying Cre was placed in the orbitofrontal cortex together with an AAV1-Flpo in the basolateral amygdala and a Cre-dependent/Flp-dependent EYFP reporter in the dorsomedial striatum. EYFP-labelled striatal neurons appeared only when both recombinase viruses were delivered, indicating that orbitofrontal axons transsynaptically label dorsomedial striatal cells, which were then recorded in slices.

BLA (Basolateral amygdalar nucleus) MBA:295 synapses onto CP (Caudoputamen) MBA:672 · Mouse

AAV1-Flpo injected into the basolateral amygdala, combined with AAV1-Cre in the orbitofrontal cortex and a double-dependent EYFP reporter in the dorsomedial striatum, produced EYFP-positive striatal neurons; no reporter signal was seen when only one recombinase virus was injected, showing that amygdalar axons transsynaptically label dorsomedial striatal neurons that also receive cortical input.
