When ChrimsonR was expressed non-specifically (CamKIIa promoter in wild-type mice, labelling both dopaminergic and GABAergic VTA cells), photo-stimulation of the VTA did not change accumbal dopamine (p = 0.0677 at 2 Hz, p = 0.416 at 20 Hz). After systemic bicuculline, the same non-specific VTA stimulation significantly increased dopamine in the NAcShell (p = 0.00452 at 2 Hz, p = 0.000243 at 20 Hz), so a functional VTA-to-NAcShell dopaminergic link exists but is masked by local GABAergic inhibition. Light delivery alone in control mice without ChrimsonR produced no change (p = 0.0923 and 0.129).

Non-specific photo-stimulation of VTA neurons, which co-activated VGAT-positive GABAergic cells (shown to express ChrimsonR in Fig. 4h), failed to raise accumbal dopamine, whereas blocking GABA-A receptors with intraperitoneal bicuculline before the identical stimulation produced a significant dopamine increase and c-fos activation of ChrimsonR-expressing dopamine neurons, indicating that local VTA GABAergic neurons inhibit VTA dopamine neurons.

In DAT-Cre mice in which ChrimsonR was restricted to dopaminergic VTA neurons, red-light stimulation through an implanted LED array in the VTA raised extracellular dopamine measured by microdialysis/HPLC in the nucleus accumbens shell; the increase was larger at 20 Hz than at 2 Hz (paired t-test p = 0.000116 at 2 Hz and p = 0.00599 at 20 Hz for the 15-min sample immediately after stimulation). Immunostaining confirmed ChrimsonR/TH co-localization and c-fos induction in these neurons after stimulation.
